In addition to sequential connectivities in the fingerprint region, sequential assignments can also be obtained in the amide region. Limited to helical areas, sequential dnn-connectivities can be observed. They can be followed from one to the next residue to establish the sequential relationship between residues, and match the connected spin system with the sequence. Note, that in this region we can trace both directions, I.e. in the beginning we don’t know whether we assign towards the C- or the N-terminus. Once the match with the sequence is found, the direction becomes unambiguous. In the above example, we can trace /connect all NOE cross peaks in this region in one contiguous stretch without a problem (starting at HN 8.52 ppm all the way to HN to 7.89. This connects 9 amide resonance frequencies, for which we have spin type assignments. Comparison of the spin type sequence with the aa sequence gives a unique match (definitely for 9 residues.)
